Panel 1: Drug Name & Indication
People say that Stacy's Clover can cure terrible viral infections, even the awful Herpes!
Acyclovir (Stacy's Clover) is an antiviral medication indicated for treating infections caused by the Herpes Simplex Virus (HSV).
Panel 2: Toxicity & Interactions
Stacy's clover likes to grow around old water pipes. If tyre-shaped 'Cyclic Spores' are also around, they can heavily damage the water filtration system!
Nephrotoxicity / Drug Interactions: Acyclovir can cause renal toxicity / acute kidney injury due to crystal precipitation in renal tubules (water pipes). Combining it with cyclosporine (cyclic spores) exacerbates nephrotoxicity.
Panel 3: Background Story
Forefathers spoke of Stacy's Clover defeating the great Herpes Dragon long ago if the dragon could ingest it.
Selective Toxicity: The drug relies on viral presence for initial activation to effectively attack the virus.
Panel 4: Administration / Uptake
The villagers took matters into their own hands and threw Stacy's Clovers at the dragon, hoping it would swallow one.
Drug Administration: Ingestion/uptake of acyclovir by infected cells.
Panel 5: Initial Activation (Monophosphorylation)
Inside the belly, the Viral TicK added a phosphate to the clover, turning it into a single-leaf clover.
Viral Thymidine Kinase (Viral TK): Acyclovir is phosphorylated by viral TK into acyclovir monophosphate inside infected cells.
Panel 6: Inactive Form
A stunned silence holds... No effect yet! Panicking begins.
Prodrug Intermediate: Acyclovir monophosphate is not yet in its fully active antiviral state.
Panels 7 & 8: Triphosphorylation
A wise old man used a slingshot to aim two special leaves into the dragon's mouth, turning it into a glowing three-leaf clover bomb.
Active Metabolite Formation: Host cellular kinases add two more phosphate groups to form acyclovir triphosphate (the active form).
Panel 9: Adverse Effect (GI Upset)
The old man started vomiting because of the toxic smell and fumes from the special leaves.
Gastrointestinal Side Effects: Common adverse effects of systemic acyclovir include nausea, vomiting, and diarrhea.
Panel 10: Viral DNA Replication Attempt
The enraged dragon started assembling his mechanical flamethrower.
Viral Polymerase Activity: The viral DNA polymerase attempts to synthesize new viral DNA using building blocks (dGTP).
Panel 11: Competitive Inhibition & Chain Termination
The three-leaf clover bomb got incorporated into the dGTP spot during assembly, halting the flamethrower completely!
Chain Termination: Acyclovir triphosphate competes with dGTP and gets incorporated into viral DNA. Lacking a 3'-OH group, it causes immediate DNA chain termination.
Panel 12: Resolution
The dragon is dead! Active infections are defeated, and Neverville lives happily with Stacy's Clover as their shield.
Therapeutic Outcome: Effective suppression and resolution of active herpes viral replication.
